Overview

Alanosine is an amino acid analogue and antibiotic derived from the bacterium Streptomyces alanosinicus. It acts as an antimetabolite with potential antineoplastic (anticancer) activity. The primary mechanism of action involves inhibition of adenylosuccinate synthetase, which converts inosine monophosphate (IMP) into adenylosuccinate—a key step in de novo purine biosynthesis. This disruption is particularly potentiated in tumors deficient in methylthioadenosine phosphorylase (MTAP), making such cancers potentially more sensitive to alanosine[1][3][5]. Clinical development has focused on its use as a targeted therapy for MTAP-deficient solid tumors such as mesothelioma, non-small cell lung cancer, soft tissue sarcoma, osteosarcoma, and pancreatic cancer[6][8]. However, clinical trials have shown limited efficacy at tested doses[8].
