Alb-ZnDDC is an albumin-based nanoparticle formulation encapsulating zinc diethyldithiocarbamate (Zn(DDC)₂), developed to enhance the stability and bioavailability of disulfiram-derived metal complexes for cancer therapy. Disulfiram, an anti-alcoholism drug, forms potent anticancer chelates with metal ions like zinc, but its clinical utility is limited by a very short plasma half-life. Alb-ZnDDC addresses these issues by providing a soluble, long-circulating delivery system. In preclinical studies, it has demonstrated significant cytotoxicity against pancreatic ductal adenocarcinoma (PDAC) across various conditions, including hypoxia and spheroid models, and has shown synergistic potential when combined with gemcitabine or paclitaxel. Research into its resistance profile has identified the cysteine protease inhibitor CST1 as a potential mediator of acquired resistance to this agent.
