Overview

Albitiazolium bromide (SAR97276) is a novel bis-quaternary ammonium compound and choline analog developed for the treatment of malaria. Its primary mechanism of action involves the inhibition of choline uptake into the *Plasmodium* parasite, which subsequently blocks the synthesis of phosphatidylcholine, a critical component of the parasite's cell membranes. By disrupting phospholipid metabolism, the drug effectively prevents parasite replication and survival. It was primarily investigated by Sanofi for both uncomplicated and severe malaria, with clinical trials exploring both parenteral and oral formulations. However, development was largely halted after Phase 2 trials.
