Albumin-bound cisplatin is a nanoparticle formulation consisting of the cytotoxic platinum-based agent cisplatin complexed with albumin. This delivery system is designed to exploit the natural accumulation of albumin in solid tumors, particularly in stroma-rich environments like pancreatic cancer, to enhance intratumor drug penetration and concentration. By utilizing albumin as a carrier, the formulation aims to improve the pharmacokinetic profile of cisplatin—increasing its plasma half-life and area under the curve (AUC)—while simultaneously reducing systemic toxicities and the volume of distribution. Preclinical studies have demonstrated that this complex significantly inhibits tumor growth and prolongs survival in various pancreatic cancer models compared to conventional free cisplatin.
