ALC-0315 is an ionizable cationic lipid that serves as a critical component of lipid nanoparticle (LNP) delivery systems for nucleic acid therapeutics. It is most notably used as one of the four lipids in the BNT162b2 (Comirnaty) mRNA vaccine developed by Pfizer and BioNTech. The lipid is designed to be neutral at physiological pH but becomes positively charged in the acidic environment of the endosome, facilitating the encapsulation of negatively charged mRNA and subsequent endosomal escape for cytoplasmic delivery. In research settings, it is frequently used as a benchmark for evaluating the efficiency and safety of novel LNP formulations.
