Overview

Alcestobart is a human IgG4 kappa monoclonal antibody targeting lymphocyte activation gene 3 protein (LAG-3, CD223), an inhibitory immune checkpoint receptor expressed on activated and exhausted T cells and other immune cells. By binding LAG-3 and blocking its interaction with major histocompatibility complex class II (MHC II) on antigen-presenting cells and tumor cells, alcestobart relieves LAG-3–mediated negative regulation of T-cell activation, enhances cytotoxic T-lymphocyte responses, and promotes antitumor immunity. Initially developed by Nanjing Leads Biolabs and now clinically co-developed with BeiGene, alcestobart is being evaluated primarily in combination with the PD-1 inhibitor tislelizumab and other agents for advanced solid tumors, including esophageal squamous cell carcinoma, non-small cell lung cancer, head and neck squamous cell carcinoma, and other malignancies.[1][2][4][5][7][13]
