Alclofenac is a nonsteroidal anti-inflammatory drug (NSAID) indicated primarily for the treatment of rheumatologic conditions such as rheumatoid arthritis, ankylosing spondylitis, and as an analgesic in painful arthritic pathologies. Its mechanism of action involves inhibition of prostaglandin G/H synthase 2 through reversible blockade of the cyclooxygenase (COX) enzyme, thereby preventing the production of inflammatory mediators like prostacyclins and prostaglandins. This results in reduced inflammation and pain[1]. Alclofenac is absorbed irregularly from the gastrointestinal tract, with maximum plasma concentrations reached within 1–4 hours after oral or rectal administration. It exhibits high protein binding (90–99%) and has a volume of distribution of 0.1 L/kg[1].
