Alda-1 is a small molecule activator of mitochondrial aldehyde dehydrogenase 2 (ALDH2). It functions as a pharmacological chaperone that increases the catalytic activity of both wild-type ALDH2 and the common inactive ALDH2*2 variant, which is prevalent in approximately 40% of East Asian populations. By enhancing ALDH2 activity, Alda-1 facilitates the detoxification of reactive aldehydes such as 4-hydroxy-2-nonenal (4-HNE) and acetaldehyde, thereby reducing oxidative stress, protecting mitochondrial integrity, and preventing cellular apoptosis. Preclinical research has demonstrated its potential therapeutic benefits in various conditions characterized by oxidative damage, including myocardial ischemia-reperfusion injury, stroke, Parkinson's disease, and major depressive disorder.
