Alda-64 is a small molecule activator of aldehyde dehydrogenase 2 (ALDH2). It is being investigated for its potential to mitigate alcohol-driven tumor progression, particularly in esophageal squamous cell carcinoma (ESCC) models with ALDH2 deficiency. ALDH2 deficiency is common in East Asian populations and leads to the accumulation of acetaldehyde, a known carcinogen. Alda-64 has been shown to increase ALDH2 expression, suppress epithelial-mesenchymal transition (EMT) markers like N-cadherin, and reduce tumor growth in vivo. It is being developed as a precision therapeutic and preventive agent for high-risk genetic and environmental contexts associated with alcohol consumption.
