Aldesleukin + interleukin-12 is an investigational combination immunotherapy regimen being evaluated by the National Cancer Institute (NCI) for the treatment of advanced solid tumors. This combination therapy pairs aldesleukin (recombinant human interleukin-2), an approved T-cell growth factor, with recombinant human interleukin-12 (rhIL-12), an experimental proinflammatory cytokine. Interleukin-12 acts by promoting the differentiation of naive T-cells into Th1 cells and stimulating the production of interferon-gamma from natural killer (NK) cells and T-cells, thereby enhancing cytotoxic activity. Aldesleukin further supports the expansion and activation of these immune effector cells. The synergy between these two cytokines is intended to overcome immune suppression within the tumor microenvironment and stimulate a robust anti-tumor response in patients with cancers such as renal cell carcinoma, breast cancer, lung cancer, and various sarcomas.
