Overview

This combination therapy consists of aldesleukin (recombinant interleukin-2), zidovudine (an NRTI), and ganciclovir (an antiviral). It was evaluated by the AIDS Malignancy Consortium (AMC) in the Phase II pilot study AMC 019 for the treatment of AIDS-related primary central nervous system lymphoma (PCNSL). Aldesleukin acts as an immunomodulatory biologic agent that functions as a T-cell growth factor to enhance immune function in profoundly immunosuppressed patients. Zidovudine and ganciclovir are included to target the underlying viral drivers of the malignancy, specifically HIV and Epstein-Barr virus (EBV), which is consistently associated with AIDS-related PCNSL. The regimen was designed as both induction and maintenance therapy to address both the immune dysfunction and the viral etiology of this rare B-cell lymphoma.
