ALDH1 siRNA is a small interfering RNA (siRNA) designed to silence the expression of Aldehyde Dehydrogenase 1 (ALDH1), a cytosolic enzyme that serves as a functional marker for cancer stem cells (CSCs) and plays a critical role in cellular detoxification and retinoic acid biosynthesis. In various malignancies, high ALDH1 activity is associated with increased tumorigenicity, chemoresistance, and poor clinical outcomes. ALDH1 siRNA acts through the RNA interference (RNAi) pathway, where it facilitates the sequence-specific degradation of ALDH1 mRNA (most commonly the ALDH1A1 or ALDH1A3 isoforms). This reduction in ALDH1 protein levels sensitizes cancer cells to chemotherapeutic agents, impairs the self-renewal and maintenance of CSC populations, and inhibits tumor growth and metastasis. While primarily utilized as a research tool in preclinical studies, ALDH1 siRNA is also being investigated in translational oncology using advanced delivery systems such as lipid nanoparticles to overcome the challenges of systemic RNA delivery.
