Overview

**Aldoxorubicin + gemcitabine** is an investigational combination therapy composed of aldoxorubicin, a tumor-targeted prodrug of doxorubicin, and gemcitabine, a nucleoside analog antimetabolite. Aldoxorubicin covalently binds to albumin and is released under acidic conditions within tumors, delivering doxorubicin to malignant tissue with reduced systemic cardiotoxicity. Gemcitabine is phosphorylated intracellularly to active metabolites that inhibit DNA synthesis, leading to cell death. This combination has been evaluated in early-phase trials for relapsed or refractory metastatic solid tumors, seeking to leverage potentially synergistic antitumor activity, with dose-limiting toxicities similar to those of the parent drugs.
