Alectos Therapeutics is developing novel small-molecule O-GlcNAcase (OGA) inhibitors as disease-modifying therapies for neurodegenerative and acute neurological diseases. The program targets the OGA enzyme; inhibition of OGA leads to increased protein O-GlcNAcylation, which helps prevent the pathogenic aggregation of proteins such as tau, alpha-synuclein, TDP-43, and amyloid-beta, while also providing neuroprotection during cellular stress. While a previous candidate, MK-8719, was developed in partnership with Merck and reached Phase 1, Alectos has since repatriated the program and is currently advancing new, optimized OGA development candidates in the preclinical stage. The program is being evaluated for Alzheimer's disease, Parkinson's disease, acute ischemic stroke, and traumatic brain injury, and includes an OGA PET ligand to measure brain target engagement.
