Alefacept is a recombinant dimeric fusion protein immunosuppressant used to treat moderate to severe chronic plaque psoriasis in adults who are candidates for systemic therapy or phototherapy. It consists of the extracellular CD2-binding portion of human leukocyte function antigen-3 (LFA-3) linked to the Fc portion of human IgG1, produced via recombinant DNA technology in Chinese Hamster Ovary cells. Alefacept works by binding to CD2 on T lymphocytes, inhibiting their activation and proliferation through blockade of the LFA-3/CD2 interaction, and by inducing apoptosis in memory-effector T lymphocytes. This results in reduced numbers and activity of pathogenic T cells involved in psoriasis pathogenesis[1][3][4][5].
