Overview

ALETA-004 is a preclinical, multi-antigen binding CAR T-cell therapy engager (CTE) developed by Aleta Biotherapeutics. It is designed to extend the cancer-killing activity of CD19 CAR T cells to treat patients with advanced acute myeloid leukemia (AML), a deadly blood cancer. ALETA-004 binds two well-known AML antigens and links them to CD19 CAR T cells using the same CD19 target protein recognized by approved anti-CD19 CAR T cell therapeutics. This approach enables AML tumor cells, which do not naturally express CD19, to be targeted and killed by existing anti-CD19 CAR T therapies by effectively changing their antigen expression profile[1][2][4]. The drug represents Aleta’s first CTE program capable of binding multiple tumor antigens and fundamentally altering tumor cell antigen expression[4].
