Overview
Alevian Duo is an approved fixed-dose combination therapy widely marketed in Latin America for the treatment of irritable bowel syndrome (IBS) and related functional gastrointestinal disorders. It combines pinaverium bromide, a locally acting, gastrointestinal-selective calcium channel blocker, with dimeticone (simethicone), a silicone-based surfactant and antifoaming agent. Pinaverium bromide selectively blocks L-type voltage-gated calcium channels in the smooth muscle of the gut, reducing hyperreactivity, relieving painful spasms, and normalizing intestinal transit without causing systemic cardiovascular side effects. Dimeticone acts physically to reduce the surface tension of gas bubbles within the gastrointestinal tract, causing them to coalesce and facilitating their elimination to relieve bloating, meteorism, and abdominal distension. Both active ingredients are poorly absorbed systemically, exerting their therapeutic effects locally within the gastrointestinal tract.
