Overview

Alexidine dihydrochloride is a bis-biguanide compound with broad-spectrum antimicrobial properties. It exhibits potent antifungal and antibiofilm activity against a diverse range of fungal pathogens—including multidrug-resistant species such as Candida auris—and has demonstrated synergy with other antifungals like fluconazole and amphotericin B. In addition to its antifungal effects, it acts as an antibacterial agent used in endodontic procedures and is structurally related to chlorhexidine. Mechanistically, alexidine dihydrochloride is an inhibitor of protein tyrosine phosphatase localized to mitochondrion 1 (PTPMT1), which may have implications for apoptosis induction and anticancer activity. Its ability to disrupt biofilms makes it a candidate for repurposing as a pan-antifungal drug[2][5][6].
