ALF43 (Army Liposome Formulation-43% Cholesterol) is a liposome-based vaccine adjuvant developed by the Walter Reed Army Institute of Research (WRAIR) and utilized in HIV vaccine research sponsored by the National Institute of Allergy and Infectious Diseases (NIAID). The formulation consists of liposomes containing a high concentration of cholesterol (43%) and typically incorporates Monophosphoryl Lipid A (MPLA) as an immunostimulant. ALF43 is designed to enhance both humoral and cellular immune responses by facilitating the delivery of antigens to antigen-presenting cells and activating Toll-like receptor 4 (TLR4). It has been evaluated in Phase 1 clinical trials, such as RV460, in combination with DNA and protein-based HIV-1 Env vaccines to improve safety and immunogenicity in healthy volunteers.
