Overview

Propofol is a sedative-hypnotic agent that acts as a positive allosteric modulator of the gamma-aminobutyric acid type A (GABA-A) receptor, producing rapid onset hypnosis and amnesia. When used together, these drugs exhibit synergistic effects, resulting in greater sedation and analgesia than either drug alone. The combination allows for lower doses of each agent, potentially reducing side effects such as nausea or prolonged recovery time. Propofol can offset some emetic effects of alfentanil, while both drugs contribute to respiratory depression and hemodynamic changes; careful monitoring is required during administration[1][4][5]. This regimen is widely utilized in outpatient procedures due to its rapid onset/offset profile.
