Alfi0894i-Fc is a novel Fc-fusion artificial lectin designed to specifically recognize and bind to high mannose (HM) linkages, which are aberrant glycan signatures found on the surface of many cancer cells, such as those in non-small cell lung cancer (NSCLC). It is derived from Alfi0894, an endoglycosidase from gut bacteria, with inactivating mutations in its glycan binding pocket to create an artificial lectin. The Fc portion of the molecule, derived from IgG1, confers stability and allows for binding to Fc receptors like FcGRIIIa, similar to Rituximab. This lectibody demonstrates high specificity and affinity for HM-expressing cells and is remarkably stable, not prone to aggregation.
