**ALFQ** is an investigational liposome-based vaccine adjuvant developed by scientists at the Walter Reed Army Institute of Research. It comprises a cholesterol-rich liposomal formulation containing the synthetic monophosphoryl lipid A analogue 3D-PHAD and the Quillaja saponaria-derived saponin QS-21. The 3D-PHAD component activates Toll-like receptor 4-mediated innate immune signaling, while QS-21 contributes to enhanced cellular and humoral vaccine responses. ALFQ is administered after mixing with a vaccine antigen and is being evaluated in early-stage intramuscular vaccine studies for HIV-1, malaria, Campylobacter jejuni infection, COVID-19, and influenza.
