Overview

ALFQ (Army Liposome Formulation containing QS-21) is a potent adjuvant system developed by the Walter Reed Army Institute of Research (WRAIR). It is a liposomal formulation that incorporates two distinct immunostimulants: 3D-PHAD (3-deacyl-monophosphoryl lipid A), a synthetic Toll-like receptor 4 (TLR4) agonist, and QS-21, a purified saponin extracted from the *Quillaja saponaria* tree. The liposomes are composed of dimyristoyl phosphatidylcholine (DMPC), dimyristoyl phosphatidylglycerol (DMPG), and cholesterol. ALFQ is designed to elicit robust and balanced immune responses, including high antibody titers and strong Th1-type cellular immunity. It has been utilized in clinical trials for various infectious disease vaccines, including those targeting *Campylobacter jejuni*, malaria (*Plasmodium falciparum*), and HIV-1, often demonstrating superior immunogenicity compared to earlier ALF-based formulations.
