ALG-001075 is a small molecule drug developed as a potent capsid assembly modulator (CAM) for the treatment of chronic hepatitis B virus (HBV) infection. It is classified as a Class II CAM, specifically targeting empty capsids, and demonstrates broad and potent antiviral activity by inhibiting HBV replication and the establishment of covalently closed circular DNA (cccDNA). In preclinical studies, ALG-001075 showed significant reductions in plasma HBV DNA levels (>5 log10 IU/mL) after extended dosing in animal models. The compound was optimized from earlier CAMs for improved potency and safety. Due to its favorable pharmacokinetic properties but limited solubility, clinical development advanced with its highly soluble prodrug form, ALG-000184. Both compounds exhibit low risk for drug-drug interactions mediated by cytochrome P450 enzymes or major transporters[1][4][5][7].
