Overview
ALG-006746 is a novel, non-heteroarylpyrimidine capsid assembly modulator (CAM-A) developed by Aligos Therapeutics for the treatment of hepatitis B virus (HBV) infection. It acts as a potent inhibitor of HBV RNA encapsidation and covalently closed circular DNA (cccDNA) establishment in infected hepatocytes. Preclinical studies have demonstrated that ALG-006746 significantly reduces HBV DNA, HBsAg, and HBeAg levels in both cell-based assays and animal models. The compound shows favorable pharmacokinetic properties with low to moderate plasma clearance and moderate volume of distribution. Its mechanism involves targeting the HBV core/capsid protein to disrupt viral replication processes essential for chronic infection[1][2][3][6].
