ALG-010133 is an S-antigen Transport Inhibiting Oligonucleotide Polymer (STOPS) molecule developed by Aligos Therapeutics for the treatment of chronic hepatitis B. It is a nucleic acid polymer (NAP), specifically a locked nucleic acid (LNA)-modified oligonucleotide, designed to inhibit the production and release of hepatitis B surface antigen (HBsAg) from infected hepatocytes with high potency in vitro. The drug acts by interacting with host cellular factors involved in RNA processing, translation, and protein folding/degradation, thereby reducing HBsAg levels. Despite promising preclinical results and acceptable safety profiles in early clinical studies, development was discontinued after Phase 1 due to insufficient efficacy at projected therapeutic doses[1][2][3][7].
