Overview

ALG-020572 is a GalNAc-conjugated antisense oligonucleotide (ASO) developed for the treatment of chronic hepatitis B. It is an 18-mer phosphorothioated ASO containing novel bridged nucleic acid/DNA modifications, designed to selectively bind to and degrade viral messenger RNA complementary to the hepatitis B surface antigen (HBsAg) region, thereby reducing HBsAg levels in patients. The drug utilizes third-generation XNA chemistry and GalNAc conjugation for enhanced hepatocyte targeting and improved pharmacological properties. Despite promising preclinical results, clinical development was terminated due to unexpected liver toxicity observed in early-phase trials[1][3][4][5][6].
