Overview

ALG-037287 is an orally bioavailable, liver-targeted small molecule inhibitor of fatty acid synthase (FASN) designed for the treatment of metabolic dysfunction-associated steatohepatitis (MASH). Developed by Aligos Therapeutics, the compound selectively targets the beta-ketoacyl reductase (KR) domain of FASN to suppress de novo lipogenesis (DNL) in the liver. By optimizing tissue distribution to achieve high liver-to-plasma concentration ratios and minimizing exposure in the skin, ALG-037287 aims to preserve therapeutic efficacy in MASH while reducing the risk of alopecia and other dermatological side effects associated with systemic FASN inhibition.
