Overview

ALG-055009 is a potent and selective small molecule agonist of the thyroid hormone receptor beta (THR-β), developed by Aligos Therapeutics for the treatment of metabolic dysfunction-associated steatohepatitis (MASH), previously known as non-alcoholic steatohepatitis (NASH). The drug acts by selectively activating THR-β in the liver, leading to reductions in liver fat and atherogenic lipids such as LDL-C, lipoprotein(a), and apolipoprotein B. Clinical studies have demonstrated significant reductions in liver fat content, favorable tolerability, and dose-dependent increases in sex hormone binding globulin (SHBG) as a marker of target engagement. ALG-055009 has shown best-in-class potential compared to other THR-β agonists due to its potency, selectivity, safety profile, and oral bioavailability[1][2][3][4][5][6][7][8].
