ALG-093702 is an orally bioavailable, liver-targeted small molecule inhibitor of programmed death-ligand 1 (PD-L1) being developed by Aligos Therapeutics. It is designed as a novel therapeutic approach for the treatment of chronic hepatitis B (CHB) and liver cancers, such as hepatocellular carcinoma (HCC). By specifically targeting the liver, ALG-093702 aims to provide the therapeutic benefits of PD-L1 blockade—restoring the activity of exhausted T cells—while potentially reducing the systemic immune-related adverse events (irAEs) often associated with traditional monoclonal antibody-based checkpoint inhibitors. Preclinical studies have demonstrated that the compound achieves high concentrations in the liver and exhibits potent anti-viral and anti-tumor efficacy in various in vivo models.
