ALG-093940 is a potent, orally bioavailable small molecule inhibitor of the PD-1/PD-L1 checkpoint pathway. Developed by Aligos Therapeutics, it distinguishes itself from traditional monoclonal antibodies by binding directly to PD-L1 and inducing its dimerization, internalization, and subsequent degradation. This mechanism of action effectively reduces PD-L1 protein levels on the surface of tumor cells, leading to enhanced T-cell activation and immune-mediated tumor cell killing. Preclinical studies have demonstrated dose-dependent tumor growth inhibition in humanized-PD-L1 mouse models and a favorable safety and pharmacokinetic profile across multiple species, supporting its potential as a once-daily oral therapy for various cancers.
