ALG-097558 is a potent, orally available small molecule inhibitor targeting the 3C-like protease (3CLpro, also known as main protease or Mpro) of coronaviruses. It demonstrates broad-spectrum antiviral activity against both alpha and beta coronaviruses, including all tested SARS-CoV-2 variants such as Omicron. In preclinical studies, ALG-097558 has shown nanomolar potency in inhibiting viral replication and is up to 20-fold more active than nirmatrelvir in cell-based assays. Its highly conserved target site suggests a high probability of retaining efficacy against future coronavirus variants. The drug was developed by Aligos Therapeutics in collaboration with KU Leuven’s Centre for Drug Design and Discovery (CD3), primarily for the treatment and prevention of COVID-19. Phase 1 clinical trials are ongoing to evaluate its safety, pharmacokinetics, and tolerability[1][2][4][5][6][7].
