ALG-125755 is a GalNAc-conjugated small interfering RNA (siRNA) developed by Aligos Therapeutics for the treatment of chronic hepatitis B. It specifically targets and inhibits the expression of hepatitis B surface antigen (HBsAg) by binding to argonaute-2 (AGO-2), which mediates RNA interference and leads to cleavage of HBV RNA in hepatocytes. Preclinical studies have demonstrated potent inhibition of HBsAg secretion with favorable safety and pharmacokinetic profiles, including a long liver half-life that may allow for less frequent dosing compared to other siRNAs. The drug is being investigated as part of efforts toward achieving a functional cure for chronic hepatitis B[1][2][5][6][7][8].
