ALG-126081 is a small interfering RNA (siRNA) therapeutic candidate developed by Aligos Therapeutics for the treatment of chronic hepatitis B virus (HBV) infection. The compound is designed to knock down the production of hepatitis B surface antigen (HBsAg) by targeting HBV RNA transcripts. It incorporates Aligos' proprietary nucleotide stabilization chemistries, specifically a 5' phosphate-mimetic end cap and 2'fluoro mimic nucleotides, which are intended to improve the potency and duration of HBsAg knockdown. Preclinical data presented at the EASL Digital International Liver Congress in 2022 demonstrated significant efficacy in AAV-HBV mouse models, showing sustained reduction of HBsAg levels.
