ALG-126101 is a small interfering RNA (siRNA) therapeutic developed by Aligos Therapeutics for the treatment of chronic hepatitis B virus (HBV) infection. The compound is designed to specifically target and knock down the production of hepatitis B surface antigen (HBsAg). It incorporates proprietary stabilization technologies, including a 5' phosphate-mimetic end cap and 2'fluoro mimic nucleotides, which are intended to enhance the potency and duration of HBsAg knockdown. Nonclinical studies in AAV-HBV mouse models have demonstrated significant improvements in viral antigen reduction compared to earlier iterations. The drug was highlighted in nonclinical data presentations at the European Association for the Study of the Liver (EASL) Digital International Liver Congress in 2022.
