Overview

ALG-170674 is a potent and selective small molecule agonist of the thyroid hormone receptor beta (THR-β), currently in preclinical development by Aligos Therapeutics. It is being investigated for the treatment of metabolic dysfunction-associated steatohepatitis (MASH), formerly known as non-alcoholic steatohepatitis (NASH). By selectively activating the THR-β isoform, which is predominantly expressed in the liver, ALG-170674 aims to enhance hepatic lipid metabolism, reduce liver fat (steatosis), and improve metabolic parameters such as LDL cholesterol and triglycerides. This selectivity is intended to avoid the adverse effects associated with the activation of the THR-α isoform, such as cardiac toxicity and bone resorption. ALG-170674 is positioned as a next-generation candidate following Aligos's lead THR-β agonist, ALG-055009.
