Drug intelligence / Profile preview
ALG-171191 is an antisense oligonucleotide (ASO) designed for the treatment of chronic hepatitis B virus (HBV) infection. Developed by Aligos Therapeutics in collaboration with Amoytop Biotech, ALG-171191 is designed to achieve a functional cure for HBV by utilizing a dual mechanism of action: RNase H-mediated direct antiviral activity (targeting HBV RNA to reduce HBsAg levels) and immunomodulatory activity via Toll-like receptor 8 (TLR8) agonism. In preclinical evaluations, ALG-171191 demonstrated enhanced in vitro RNase H-mediated activity and deeper HBsAg knockdown in mouse models compared to clinical-stage benchmarks like bepirovirsen. However, during lead optimization, sibling candidates ALG-170675 and ALG-171036 were selected as the optimal candidates for further development due to superior safety profiles in HBV-infected humanized liver mouse models.
