Drug intelligence / Profile preview
ALG-HSD #1 is a preclinical GalNAc-conjugated small interfering RNA (siRNA) candidate designed to selectively silence the expression of 17-beta-hydroxysteroid dehydrogenase 13 (HSD17B13) in hepatocytes. Developed by Aligos Therapeutics in collaboration with Merck & Co., the therapeutic is being investigated for the treatment of metabolic dysfunction-associated steatotic liver disease (MASLD) and metabolic dysfunction-associated steatohepatitis (MASH). By targeting HSD17B13, a liver-enriched lipid droplet-associated protein linked to liver inflammation and injury, ALG-HSD #1 aims to mimic the protective effects of loss-of-function genetic variants. In preclinical models, including non-human primates, the candidate has demonstrated robust and durable knockdown of HSD17B13 mRNA and protein levels with a favorable safety profile.
