Overview

ALG.APV-527 is a bispecific agonistic antibody designed to target the co-stimulatory receptor 4-1BB (CD137) and the tumor-associated antigen 5T4. It selectively activates T cells and natural killer (NK) cells only upon simultaneous binding to both targets, which are preferentially expressed in tumor tissues such as triple negative breast cancer and renal cell carcinoma. This conditional activation aims to stimulate tumor-specific immune responses while minimizing systemic immune activation and associated toxicities like liver toxicity seen with other 4-1BB targeting agents. The molecule was developed using Alligator Bioscience’s proprietary ALLIGATOR-GOLD® antibody library combined with Aptevo Therapeutics’ ADAPTIR™ technology platform. It is currently under clinical investigation primarily for advanced solid tumors.
