Overview

ALIA-1758 is a bispecific monoclonal antibody developed for the treatment of Alzheimer's disease. It targets pyroglutamate amyloid beta (3pE-Aβ), a form of amyloid beta that aggregates in plaques, and incorporates a transferrin receptor (TfR) binding module to facilitate efficient delivery across the blood-brain barrier. The antibody is designed to clear amyloid plaques from the brain with high specificity and efficiency, potentially at lower doses than other anti-amyloid therapies. Its optimized Fc domain increases half-life and reduces pro-inflammatory cytokine release, aiming to minimize immune-related side effects such as ARIA. ALIA-1758 was originally developed by Aliada Therapeutics using their MODEL technology platform and has advanced into Phase 1 clinical trials for Alzheimer's disease. In October 2024, Aliada Therapeutics was acquired by AbbVie, which now leads its development[1][2][3][4][5][6][7][8].
