Overview

Alintegimod is a first-in-class, orally bioavailable small molecule that acts as an allosteric activator of the integrin cell adhesion receptors very late antigen-4 (VLA-4; integrin alpha4/beta1) and lymphocyte function-associated antigen-1 (LFA-1; integrin alphaL/beta2). By selectively activating these integrins, alintegimod augments T cell activation and trafficking, enhances immune cell interactions required for effective immune responses, and may improve the efficacy of cancer immunotherapies—particularly in patients resistant to immune checkpoint inhibitors. It is being developed primarily for use in combination with other immunotherapies such as ipilimumab and nivolumab. The drug is administered orally as a soft gel capsule. Alintegimod is under clinical development by 7 Hills Pharma for various solid tumors and other indications including chagas disease, melanoma, pleural mesothelioma, renal cell carcinoma, MSI-high or mismatch repair-deficient colorectal cancer, hepatocellular carcinoma, non-small cell lung cancer, and was previously considered for influenza[1][2][4][5][6][8].
