Overview

Alisertib + sapanisertib is an investigational combination therapy consisting of two small molecule inhibitors targeting distinct oncogenic pathways. Alisertib is an Aurora A kinase inhibitor that disrupts mitotic spindle formation and cell division, while sapanisertib (MLN0128) is a dual mTORC1/2 inhibitor that blocks the mammalian target of rapamycin pathway involved in cell growth and survival. The combination has been evaluated in phase 1b/2 clinical trials for patients with advanced or refractory solid tumors, including pancreatic adenocarcinoma. Preclinical studies suggested potential synergy by overcoming resistance mechanisms associated with monotherapy. Clinical results indicate modest activity overall but notable responses in select patients; toxicity was consistent with known profiles of each agent[1][5].
