The ALK-mRNA Vaccine is an investigational messenger RNA (mRNA) vaccine designed to target anaplastic lymphoma kinase (ALK)-driven malignancies. It consists of a codon-optimized mRNA encoding the cytoplasmic domain (exons 20–29) of the ALK oncoantigen, formulated with lipid nanoparticles (LNPs). The vaccine aims to induce a strong, ALK-specific cytotoxic T-cell immune response to combat ALK-positive tumors, addressing challenges like resistance and relapse seen with tyrosine kinase inhibitors. Preclinical studies in mouse models have demonstrated its ability to efficiently express ALK, present immunogenic peptides, and elicit a potent, non-toxic CD8+ ALK-specific T-cell response, outperforming peptide vaccines. These results support its further investigation in clinical trials.
