Overview

ALK-specific TCR-T cells are an adoptive cell therapy consisting of T cells engineered to express a T-cell receptor (TCR) specific for a peptide derived from the Anaplastic Lymphoma Kinase (ALK) protein. Specifically, these cells target the ALK-derived peptide RPRPSQPSSL presented by the HLA-B*07:02 MHC class I molecule. ALK is a known oncogenic driver in several cancers, including anaplastic large cell lymphoma (ALCL), non-small cell lung cancer (NSCLC), and neuroblastoma. By targeting a naturally presented intracellular peptide, these TCR-T cells can recognize and eliminate ALK-expressing tumor cells, including those resistant to ALK tyrosine kinase inhibitors like crizotinib. The therapy has demonstrated potent anti-tumor activity in preclinical models, both as a monotherapy and in combination with ALK inhibitors like crizotinib, which has been shown to further enhance tumor regression and survival.
