ALK-Vac is a prophylactic peptide vaccine designed to target common resistance mutations in the ALK kinase domain of patients with advanced ALK-positive non-small cell lung cancer (NSCLC). Developed by researchers at the Johns Hopkins University School of Medicine, the vaccine consists of synthetic long peptides targeting seven specific ALK resistance mutations (I1171T, I1171N, I1171S, L1196M, G1202R, D1203N, and E1210K) combined with the poly-ICLC adjuvant. It is intended for use in patients who are currently responding to standard-of-care tyrosine kinase inhibitor (TKI) therapy to delay or prevent the emergence of resistance. In the Phase 1b ARCHER trial, ALK-Vac demonstrated safety and the ability to induce vaccine-specific T-cell responses against multiple resistance-associated antigens.
