ALK101sc is an afucosylated IgG1 bispecific antibody targeting the epidermal growth factor receptor (EGFR) and the hepatocyte growth factor receptor (c-MET), co-formulated with hyaluronidase for subcutaneous administration. Developed by Allink Biotherapeutics using a single-cell common-light-chain (CLC) platform, it is designed to overcome resistance to EGFR tyrosine kinase inhibitors (TKIs) in non-small cell lung cancer (NSCLC). The drug functions by binding to both receptors, inducing their internalization and lysosomal degradation, and inhibiting downstream ERK and AKT signaling. Its afucosylated Fc region further enhances antitumor activity through antibody-dependent cellular cytotoxicity (ADCC). Preclinical studies indicate that the subcutaneous formulation of ALK101sc offers comparable systemic exposure to intravenous amivantamab while potentially reducing infusion-related reactions and improving patient convenience.
