Drug intelligence / Profile preview
ALK4-Fc is a recombinant soluble fusion protein consisting of the extracellular domain of activin receptor-like kinase 4 (ALK4, also known as ACVR1B) fused to the Fc domain of an immunoglobulin G (IgG). It functions as a ligand trap that binds and neutralizes Smad2/3-activating ligands of the TGF-beta superfamily, including Activin A, GDF8 (Myostatin), and GDF11. By sequestering these ligands, ALK4-Fc prevents their binding to endogenous type I and type II receptors, thereby inhibiting downstream Smad2/3 phosphorylation and signaling. In preclinical research, ALK4-Fc has been investigated for its potential to promote osteoblast differentiation, increase bone mineral density, and preserve muscle mass, making it a valuable tool for studying therapeutic approaches to osteoporosis, sarcopenia, and neuromuscular disorders.
