Drug intelligence / Profile preview
"ALK7 RNAi" is an investigational, preclinical-stage small interfering RNA (siRNA) therapy developed by Rona Therapeutics and licensed globally to Roche. The candidate utilizes Rona's proprietary extrahepatic delivery platform (APOLLO) to target activin receptor-like kinase 7 (ALK7), a type I receptor in the transforming growth factor-beta (TGF-β) superfamily encoded by the *ACVR1C* gene. ALK7 is predominantly expressed in adipose tissue, where its signaling pathway regulates lipid storage, lipolysis, and energy homeostasis. By silencing ALK7 directly in adipocytes, the therapy aims to reduce visceral and ectopic fat deposits while preserving lean mass. This mechanism is distinct from and potentially complementary to central nervous system-acting, appetite-regulating incretin-based therapies. In October 2026, Roche acquired an exclusive worldwide license to develop and commercialize the candidate for obesity and metabolic diseases in a deal valued at up to $1.16 billion, including $100 million in upfront and near-term payments.
