Overview

ALKS 6610 is a novel, orally administered, peripherally-acting mu-opioid receptor antagonist (PAMORA) developed by Alkermes. It was primarily investigated for the treatment of opioid-induced constipation (OIC), a common side effect in patients receiving opioid therapy for chronic pain. The compound is designed to selectively antagonize mu-opioid receptors in the gastrointestinal tract to restore bowel function without crossing the blood-brain barrier, thereby preserving the central analgesic efficacy of the opioids. Clinical development included a Phase 1 single ascending dose study in healthy adults to assess safety, tolerability, and pharmacokinetics, including the impact of food on its absorption. However, the program was subsequently discontinued from the Alkermes pipeline and is no longer in active development.
