Overview

ALL-001 is an investigational oromucosal immunotherapy being developed by Allero Therapeutics for the treatment of celiac disease. The drug is based on the company's proprietary Specific Immune Tolerance Induction (SITI) platform, which utilizes a blend of gluten-derived peptides delivered to the oral mucosa. The mechanism of action involves the recruitment and activation of tolerogenic dendritic cells in the oral cavity, which subsequently induce the differentiation and expansion of gluten-specific regulatory T cells (Tregs). These Tregs are intended to migrate to the gut and suppress the inflammatory T-cell response to gluten, thereby restoring immune tolerance and preventing intestinal mucosal damage. As of 2024, ALL-001 is in the IND-enabling phase of development.
